Mouse/Rat Adult – Brain (Cerebellum)
Electrode Recommendation:
For gene transfection into Adult Rat or Mouse Brain (cerebellum) by electroporation we recommend the CUY200S electrode set and the CUY21SC electroporator.
The proposed CUY21SC and CUY200S solution does not require as many components (as other solutions) and employs a simpler methodology.
The CUY200S electrode set is a pair of needle electrodes. The inside of the needles is hollow, so they can be used for injection of the plasmid. Accordingly, electroporation may be performed immediately after injecting the plasmid.
Alternatively, one could use the CUY567 electrode. It too is a needle electrodes but it cannot be used to inject plasmid. Accordingly, one must first inject genes into the brain and then insert the needle electrodes into the target where the plasmid is injected. The CUY200S is a much more appropriate and elegant solution for this application, hence why we recommend the CUY200S.
The CUY21SC electroporator was specifically designed for in vitro applications that require the delivery of genes into very sensitive targets e.g., adult mouse’s brain and single cells. In such applications, delivery of a controlled and measurable low current (<10 mA) is essential in order to ensure no damage is done to the target. The CUY21 SC model enables the researcher to measure the delivered (low) current.
The optimal current for this type of application is 4-6mA. If the actual current is higher than 8mA, it will cause serious damage to the target area of the brain. If the current is lower than 5mA, transfection efficiency will be very low. Thus, being able to check and control the actual current is a key component for successful transfection in this application. The CUY21 SC electroporator has been specifically designed to enable the researcher to measure and control currents lower than 10mA. Accordingly, unlike other solution packages one does not require an additional miliamper meter (oscilloscope). Hence, why we recommend the CUY21SC for such applications.
